On the settings page it says:
Lineups Change Daily - Slots Lock Individually at Scheduled Gametime
and
Roster Locktime Daily - Rosters Lock at First Game of Day
Does this mean, if there's a day game, that we can't change our rosters for later games after the day game starts?

Okay, these refer to two different things. You can move a player on your roster until that particular player's game starts, but once the first game of the day begins, you can no longer add or drop a player for that day's games.

Lineups - players already on your team. You can switch them from the bench to starting spots as the games and times indicate.
Rosters - FA or WW pick-ups. If you add somebody after the first game is played, that player will show up on your roster for the NEXT day, not the day you add said player.

Reddads league last year was if there was a 1:00pm EST game after that game started you could not make any moves on your roster that day. While other leagues the 1:00pm games were locked while other games you were allowed to make moves

I like the idea of being able to pick up a player at any time. Can we make it so you can pick up a player and they just get added the next day? Otherwise I have a feeling that we would have to wait until 3am EST to add players.

You can pick up a player whenever you want to, but if you do it after the first game of the day, he will show up on the next day's roster. If it's Wednesday, and the games have already started when you pick a player up, just click the tab that says Thursday and the player you picked up will be there.
